聲光可調(diào)—近紅外光譜法快速測定丹參藥材中隱丹參酮的含量

          摘 要 目的:建立快速測定丹參藥材中隱丹參酮含量的方法。方法:采用高效液相色譜法測定藥材樣品中隱丹參酮含量(作為參考值)。采用聲光可調(diào)-近紅外光譜法結(jié)合偏最小二乘法建立藥材樣品中隱丹參酮含量的定量模型:根據(jù)藥材樣品中隱丹參酮含量測定結(jié)果,采集35份藥材樣品,以一階導(dǎo)數(shù)法聯(lián)合平滑濾波系數(shù)法預(yù)處理光譜,藥材樣品中隱丹參酮含量測定最佳波段范圍為1 250~2 150 nm。結(jié)果:藥材樣品中隱丹參酮含量測定方法學(xué)經(jīng)驗證符合要求。隱丹參酮定量模型的校正均方根偏差為0.014 6,預(yù)測均方根偏差為0.022 3,相關(guān)系數(shù)為0.976 6。內(nèi)部驗證偏差為2.41%,外部驗證偏差為4.06%。結(jié)論:該方法快速準(zhǔn)確,簡便無污染,可用于丹參藥材中隱丹參酮含量的快速測定。

          ABSTRACT OBJECTIVE: To establish rapid method for content determination of cryptotanshinone in Salvia miltiorrhiza. METHODS: The content of cryptotanshinone in sample was determined by HPLC (as reference value). AOTF-NIDRS combined with PLS was used to establish quantitative correction model for the content of cryptotanshinone in S. miltiorrhiza. According to the results of content determination of cryptotanshinone in samples, 35 samples of medicinal material were collected. First-order derivative combined with smoothing filter coefficient method was used to pretreat spectrum, and optimal band range for content determination of cryptotanshinone in sample ranged 1 250-2 150 nm. RESULTS: Methodology validation of content determination of cryptotanshinone in sample was in line with the requirements. Correction mean square deviation of quantitative correction model of cryptotanshinone was 0.014 6, and predicted mean square deviation was 0.022 3, coefficient of association was 0.976 6. The internal verification deviation was 2.41% and the external verification deviation was 4.06%. CONCLUSIONS: This method is rapid, accurate, simple and pollution-free. It can be used for rapid content determination of cryptotanshinone in S. miltiorrhiza.

          丹參為唇形科植物丹參(Salvia miltiorrhiza Bge.)的干燥根及根莖,有活血祛瘀、養(yǎng)血安神、涼血清心的功效,系應(yīng)用歷史悠久的常用中藥[1]。隱丹參酮是丹參的主要脂溶性提取物,有研究證實其在心血管疾病、代謝紊亂性疾病及神經(jīng)退行性疾病等方面有良好的應(yīng)用前景[2],而丹參藥材資源分布遍及全國,其質(zhì)量參差不齊。目前,藥品生產(chǎn)單位按2015年版《中國藥典》(一部)標(biāo)準(zhǔn)對丹參藥材進行質(zhì)量控制,但步驟復(fù)雜,檢測效率較低。因此,建立一種快速有效的檢測方法對丹參藥材質(zhì)量控制具有重要意義。
          近紅外光譜(Near infrared reflectance spctroscopy,NIR)檢測技術(shù)是近年來發(fā)展迅速的一種綠色分析技術(shù)[3],第5代聲光可調(diào)(Acousto-optic tunable,AOTF)-NIR儀具有體積小、便于攜帶、性能穩(wěn)定,環(huán)境適應(yīng)性強等特點[4],可用于在線生產(chǎn)和室外現(xiàn)場。本研究采用AOTF-NIR技術(shù)[5-6],建立快速測定丹參藥材中隱丹參酮含量的新方法,以期為丹參藥材的快速定量分析提供參考。
          1 材料
          1.1 儀器
          AOTF便攜式Luminar-5030型NIR儀,包括石英檢測皿、樣品旋轉(zhuǎn)器、SNAP光譜分析軟件、CAMO化學(xué)計量學(xué)軟件(美國Brimrose公司);Thermo-U3000型高效液相色譜(HPLC)儀,包括紫外檢測器、Chromeleon7色譜工作站(美國Thermo Scientific公司);ME204E型萬分之一電子分析天平、MS105DU型十萬分之一電子分析天平(美國Mettler-Toledo公司);SB-1200D型超聲波清洗器(寧波新芝生物科技股份有限公司)。
