高效液相色譜定量分析比較4種蟲草藥材的核苷類成分

[摘要] 為比較4種常見蟲草類藥材(冬蟲夏草、蛹蟲草、蟲草菌絲體和蟬花)中核苷類成分的差異,該研究以0.5%磷酸溶液提取樣本,色譜柱為Zorbax SB-Aq(4.6 mm×150 mm,5 μm),流動相為0.04 mol·L-1磷酸二氫鉀溶液(A)和乙腈(B),梯度洗脫,流速為0.8 mL·min-1,檢測波長260 nm,柱溫30 ℃,對4類藥材產(chǎn)品進(jìn)行分析。已知核苷以外標(biāo)一點法進(jìn)行計算,未知成分以腺苷為對照進(jìn)行計算,并對測定結(jié)果進(jìn)行聚類分析,同時利用指紋圖譜相似度軟件對不同樣本進(jìn)行了比較。結(jié)果顯示,冬蟲夏草、蛹蟲草、蟲草菌絲體、蟬花4種藥材中的核苷類成分差異較大,聚類分析和指紋圖譜分析均可有效區(qū)分冬蟲夏草和其他3種蟲草藥材。該研究建立的核苷測定方法準(zhǔn)確可靠,可用于蟲草類產(chǎn)品的品質(zhì)評價。

[Abstract] To compare the main nucleosides in Cordyceps genus herbs (C. sinensis, C. millitaris, Hirsutella sinensis and C. sobolifera), an HPLC method for simultaneous determination of uridine, inosine, guanosine, adenosine and cordycepine in Cordyceps genus herbs was developed. The sample was extracted with 0.5% phosphoric acid solution to prepare test solution. The separation was performed on a Zorbax SB-Aq (4.6 mm×150 mm, 5 μm) column with gradient elution by 0.04 mol·L-1 potassium dihydrogen phosphate solution and acetonitrile, column temperature 30 ℃,flow rate 0.8 mL·min-1,and detection wavelength 260 nm. The content of nucleosides in four Cordyceps genus herbs was evaluated by fingerprint analysis and hierarchical cluster analysis (HCA). The calibration curves of five nucleosides showed good linear regression (r>0.99) and the average recoveries were between 95.0% and 105.0%. The contents of the five nucleosides in the four Cordyceps genus herbs were different and could be obviously distinguished by HCA. The fingerprint analysis result showed that the similarity between C. sinensis and the others was less than 0.9. The method was accurate and reliable, which can be used for quality control of Cordyceps genus herbs.

冬蟲夏草Cordyceps sinensis為傳統(tǒng)名貴中藥材,具有免疫調(diào)節(jié)、抗腫瘤、抗衰老等功效[1-3]。由于其資源稀缺,無法滿足廣大消費者的需求,市場上出現(xiàn)了多種類似藥材,其中蛹蟲草C. militaris、蟲草菌絲體Hirsutella sinensis和蟬花C. sobolifera為3種最為常見的蟲草類藥材[4-7]。冬蟲夏草為冬蟲夏草真菌寄生在蝙蝠蛾科昆蟲幼蟲上形成的子座和幼蟲尸體的干燥復(fù)合體[8];蛹蟲草是蛹草真菌以蟲蛹為寄主或在人工培養(yǎng)基上培養(yǎng)形成的子實體[9];蟲草菌絲體為中華被毛孢真菌通過液體深層培養(yǎng)技術(shù)生產(chǎn)的菌絲體[10];蟬花為蟬花真菌寄生于蟬類若蟲形成的菌蟲復(fù)合體[11]。為了比較蟲草類產(chǎn)品的品質(zhì),本研究室前期發(fā)表了高效液相色譜質(zhì)譜聯(lián)用方法對冬蟲夏草、蛹蟲草和蟲草菌絲體的定性分析研究,結(jié)果顯示尿苷、肌苷、鳥苷、腺苷和蟲草素為蟲草類藥材的主要核苷類成分[12-13]。本研究在前期研究基礎(chǔ)上采用高效液相色譜方法定量分析了4種蟲草類藥材的核苷類成分,分別對15批冬蟲夏草藥材、7批蛹蟲草藥材、5批蟲草菌絲體藥材和4批蟬花藥材中的主要核苷類成分進(jìn)行了含量測定,并用聚類分析對這些藥材的核苷含量數(shù)據(jù)進(jìn)行了分類,運用相似度比較軟件對核苷類指紋圖譜進(jìn)行了比較。
1 材料
Agilent1260高效液相色譜儀(在線脫氣機,四元泵,自動進(jìn)樣器,柱溫箱,DAD檢測器);Agilent6530四級桿飛行時間質(zhì)譜檢測器(QTOF);XS205DR型天平(梅特勒-托利多儀器有限公司);KQ-500DB超聲清洗儀(昆山市超聲儀器有限公司)。
