基于DNA條形碼(ITS2)的中藥材防風與其混偽品的鑒定

[摘要] 目的 通過ITS2序列分析,對防風及其易混偽品進行鑒定,確保用藥安全。 方法 從GenBank數(shù)據(jù)庫下載中藥材防風與其混偽品的DNA條形碼ITS2序列,利用MEGA 6.06軟件進行比對分析,計算GC含量及變異位點、種內、種間遺傳距離,并構建NJ系統(tǒng)發(fā)育樹,應用ITS2數(shù)據(jù)庫網(wǎng)站預測其ITS2二級結構。 結果 序列分析與遺傳距離結果顯示,防風與其混偽品存在明顯差異,基于ITS2序列的NJ樹及其ITS2二級結構均可以將防風與其混偽品進行區(qū)分。 結論 ITS2能夠準確鑒定防風及其易混偽品,可用于中藥材的快速鑒別。

[Abstract] Objective To identify Saposhnikovia divaricata and its adulterants by using ITS2 sequence analysis and to secure medication safety. Methods ITS2 sequences of Saposhnikovia divaricata and its adulterants were downloaded from GenBank,MEGA 6.06 software was used for sequence analysis to summarize GC content,variation sites,estimate intra-and inter-specific genetic distances,and built NJ trees.Moreover,the secondary structure of ITS2 was predicted by using the ITS2 database websites. Results ITS2 sequence analysis and genetic distances calculation showed significant difference in the samples.Further more,NJ tree and secondary structure results could distinctly differentiate quality product and adulterants. Conclusion ITS2 can effectively identify Saposhnikovia divaricata and its adulterants,as a result,it can be used for rapid identification of Chinese medicinal materials.

防風(Saposhnikovia divaricata)是雙子葉植物傘形科(Umbelliferae)多年生草本植物防風屬(Saposhnikovia)防風的干燥根[1],是常用的中藥材之一,主要產于我國北部和東北部,分布于東北、華北及陜西、寧夏、甘肅、山東等地[2]。防風具有祛風解表、除濕止痛、疏肝解痙、殺蟲止癢的功效,主治外感風寒,頭痛項強,目;铦,風寒濕痹,骨節(jié)酸痛,腹痛泄瀉,腸風下血,破傷中風,麻疹難透,風疹瘙癢,瘡瘍初起。由于防風的藥用價值高,市場需求極大。近年來,部分不法商人以川防風、杏葉防風、華中前胡、田葛縷子、硬阿魏、野蘿卜等冒充正品防風,影響了用藥療效及安全。傳統(tǒng)的防風鑒定方法主要是從基原、性狀、顯微、理化等方面進行區(qū)別[3-5],這些方法對操作人員的專業(yè)知識要求較高,具有一定的局限性,難以滿足中醫(yī)藥現(xiàn)代化發(fā)展的要求,因此,亟需建立一種簡單、快速、準確的鑒定防風藥材及其混偽品的方法。
DNA條形碼[6](DNA barcoding)技術是用一段標準的、較短的DNA序列作為物種標記的一種物種分子鑒定方法,可以對生物物種實現(xiàn)準確、快速、簡便的鑒定!癉NA條形碼”首先由加拿大動物學家、皇家學會會員Paul Hebert于2003年提出,是在分子分類學和DNA分類學的基礎上用單一的小片段基因作為物種的條形碼,進而為整個生物界的物種編碼,達到類似超市利用商品條形碼可以快速準確識別成千上萬種商品的效果[7]。
